Eine der Hauptursachen fur die Entstehung einer Herzinsuffizienz ist die dilatative Kardiomyopathie (DCM), die als Herzmuskelerkrankung durch eine Erweiterung der Herzkammern und eine systolische Funktionsstoerung charakterisiert ist. Aufgrund der Vielzahl an Genen und Genvarianten, die mittlerweile als DCM-assoziiert identifiziert werden konnten, ist es interessant die Wechselwirkung verschiedener Gene genauer zu untersuchen. In dieser Arbeit wird die Rolle von NEXN genauer betrachtet und in Zusammenhang mit der Expression von unterschiedlichen Genen des Kalziumhaushaltes gesetzt. NEXN wurde erstmals 2009 von der Arbeitsgruppe um Hassel und anderen in einen Zusammenhang mit der DCM gebracht. Spater wurde die Rolle von Nexilin mittels eines konventionellen Knock-outs im Mausmodell genauer untersucht, fuhrte aber zu einer sehr fruhen Sterblichkeit der Mause. Um dem humanen Krankheitsbild naher zu kommen, wurden in dieser Arbeit Mause untersucht, in denen der konditionelle Cre-loxP Knock-out von NEXN spezifisch nur in den Kardiomyozyten erfolgt war. Zunachst wurde die verringerte Expression von Nexilin in diesen Mausen im Vergleich zum Wildtyp mittels Western Blot nachgewiesen. Dann wurde mittels verschiedener Farbungen von Herzschnitten die Entwicklung einer DCM mit fortschreitendem Alter durch den Vergleich von 12 und 56 Wochen alten Mausen betrachtet.
